Infinity Pharma halts pancreatic cancer trial

Infinity Pharmaceuticals Inc. has voluntarily halted a Phase 2 study of its drug target to treat metastatic pancreatic cancer, after early results indicated the goal endpoint of overall survival would not be met.

The Cambridge company develops IPI-926 (also known as saridegib) as a small-molecule inhibitor of Smoothened, a key component in the Hedgehog pathway, which operates only in the presence of cancer, signaling tumors cells and spreading the cancer. Infinity was testing IPI-926 with gemcitabine, in comparison to placebo with gemcitabine. The IPI-926 combination showed a higher disease rate than the placebo combination, with Infinity’s combination indicating a median survival rate less than the six-month median of gemcitabine alone.

Analysis results are not yet completed.

Infinity is also testing IPI-926 in Phase 2 trials as a potential treatment for myelofibrosis, a bone marrow malignancy that currently has no cure, and chondrosarcoma, a rare cartilage cancer that can be life threatening. The myelofibrosis trial results are expected in the second half of 2012. Results of the trial for chondrosarcoma, for which Infinity got an FDA-designated orphan drug status in March,  are due in the first half of 2013.

Infinity’s IPI-926 is the first drug candidate coming out of a $75 million alliance with Purdue Pharmaceutical Products LP of Stamford, Conn., and Mundipharma International Corp. Ltd. of the UK.
